Derbyshire County Council closed a section of the A6 due to safety concerns of trees potentially falling on to the road.
The authority announced the decision to close part of the road in Buxton to all traffic and pedestrians at 19:44 GMT on Friday.
The council said the concerns were about trees on land next to the A6 and it was in the process of organising contractors to remove them.
Barriers have been placed after the roundabout on Bakewell Road, near Morrisons, and just before the roundabout leading to Fairfield Road, near Weatherspoons, said the council.
Diversions are now in place and the council said: "We do realise that this will cause issues with traffic in the town, but public safety must come first."
