Those eligible are people aged 75 years and over, those in care homes and those aged six months and over with a weakened immune system.
People will be invited for their booster, their GP may offer them the vaccine or they can book using the NHS app for Apple or Android.
Those eligible can also find their nearest walk-in vaccination site from the NHS website., external
The national booking system is for public clinics for appointments up to 30 June.
The Royal Devon University Hospitals NHS Foundation Trust online vaccination clinic calendar, external has the most up to date clinic dates.
Leigh Mansfield, associate director of operations who is leading the vaccination programme for Royal Devon, said: "I would urge anyone who is eligible but has yet to book an appointment to not delay, and get protected against the virus as soon as possible.
"We are running clinics at sites across the region, so it’s easy and convenient to book at a time and place to suit you.
"Walk-in vaccinations are also available at all sites.
"Covid-19 can still be very dangerous to older people and those who have weakened immune systems, and this dose will top-up previous protection, reducing the risk of serious illness."
