Officers were called at 17:09 GMT on Friday to reports that a car, believed to be a white Seat Leon, had hit a pedestrian on Manford Way, Hainault.
The woman, 48, was pronounced dead at the scene after paramedics and the London Air Ambulance arrived, the force said.
The driver of the car failed to stop and the Seat Leon was found nearby.
