Michael Thornely's second championship century of the summer gave Leicestershire hope of saving their clash with Essex at Chelmsford.
The visitors resumed on 32-2, needing 253 to avoid the follow-on.
But Thornely put on 178 for the fourth wicket with Matthew Boyce (77) before being dismissed for 115 by David Masters, who finished with 6-47.
Shiv Thakor also hit an unbeaten 53 as Leicestershire reached the close on 336-9, 98 behind but with one day left.
