Glamorgan skittled Lancashire for 123 before James Anderson took 3-28 to give his side some sort of foothold.
The visitors' batsmen never got going, Michael Hogan bowling Paul Horton for a duck in the first over of the day.
Seamers Hogan and John Glover took three wickets each as just three batters got into double-figures.
The hosts reached stumps 110-4 and it could have been worse for Lancashire had Simon Kerrigan and Anderson not grabbed a wicket apiece late on.
