Hampshire seamer David Balcombe took five wickets but Andrea Agathangelou led a Lancashire fightback with a maiden half century at Southampton.
Agathangelou had been called up as a late replacement for Simon Katich, who was taken to hospital after being hit by a ball on the head in the nets.
Stephen Moore and Michael Proctor put on a first-wicket stand of 54 before Balcombe claimed 5-92.
But Agathangelou's unbeaten 93 saw Lancashire close on 252-9.

"An awful second over this morning cost David Balcombe 21 runs but he bounced back well for his eighth five-wicket haul of his career.
"It's a slow track even though all nine Lancashire batsmen fell to catches behind the wicket.
"It was a frustrating end to the day for Hampshire as the last two wickets have put on 70 runs so far."
