Wayne Madsen hit his second first-class century of the season as Derbyshire piled on the runs against Kent.
Matt Coles (4-75) reduced the hosts to 35-3 in Derby as he trapped Chesney Hughes lbw, bowled Hamish Rutherford and had Billy Godleman caught at gully.
Madsen and Neil Broom then shared a 161-run fourth wicket stand, before Madsen fell to James Tredwell for 103.
Jake Ball trapped Broom in front for 96 but Shiv Thakor's unbeaten 86 helped Derbyshire close on 381-7.
