Hampshire picked up maximum batting points for the first time this season as they took control against Durham.
They added 153 to their overnight score of 319-6 before declaring on 472-9 after lunch on the second day.
Sean Ervine was last man out with 93 from 153 balls, after sharing an eighth-wicket partnership of 95 with all-rounder Gareth Berg (56).
Ryan McLaren claimed two wickets as Durham reached 79-2 before rain brought play to an end at Chester-le-Street.
McLaren's victims included opener Mark Stoneman, who Hampshire confirmed before the start of play they have made a 28-day approach to speak to with a view to a possible move next season.
England hopeful Scott Borthwick was the South African's other wicket, having failed to press his case for a Test place in the forthcoming series against Pakistan, scoring just five.
Earlier, Durham struggled without the services of opening bowler Chris Rushworth, who was unable to bowl following a hamstring strain sustained on the first day.
Four bowlers shared two wickets each, including Keaton Jennings with his right-arm medium pacers.
