Craig Meschede's 87 helped Glamorgan recover to 294 all out after the Sussex seamers dominated the first half of the opening day.
Sussex closed on 79-3 after a frenetic final hour at Colwyn Bay.
The prolific Jofra Archer (3-67) made early breakthroughs for Sussex, while Ollie Robinson (4-46) and Chris Jordan (3-82) shared the other wickets.
Earlier Glamorgan slipped to 148-6 despite Nick Selman's patient 58 and Kiran Carlson's lively 47.
It was a day of fluctuating fortunes with Ben Brown's decision to field first vindicated by his bowlers against a raw Glamorgan line-up.
Archer passed 50 Championship wickets for the season, the first man to do so, before Meschede shared a stand of 106 for the seventh wicket with Tom Cullen (42).
Promotion-chasing Sussex then started with a 50 stand between Angus Robson (44 not out) and Luke Wells, before losing three wickets in the last 10 overs against a Glamorgan side resting five senior players ahead of T20 Blast Finals Day.
Glamorgan all-rounder Craig Meschede told BBC Wales Sport:
"We came back nicely after not the greatest of starts, Selman and Carlson had a good partnership before myself and Cullen managed to build again although we were still slightly below par.
"Again, we didn't get off to the best of starts with the ball but we came back nicely with a bit of nibble in the pitch. There's enough in the wicket, but it'll be crucial to be consistent with the new ball.
"It's the first time I've been part of the senior football side (for warm-ups) at 25, but it's a great opportunity for the young guys to showcase their skills."
Sussex bowler Ollie Robinson told BBC Sussex:
"It was great, really chuffed with how the day's gone after getting back fit and in the first team.
"I had an operation in November on my calves but there were complications so I had a second one which delayed me coming back even longer, so I haven't been playing much of the season. It's taken me a long time to get back but it's been worth it.
"The wicket started off a bit slow and it nipped around a bit, but it seems to have flattened out with the odd one doing a bit, hopefully we can get a lead and we'll be right in the mix."
