Hoffenheim left-back Fabian Johnson volleyed home the hosts' opener before ex-Fulham forward Clint Dempsey poked home their second after the break.
Turkey, who failed to qualify for this summer's finals, scored a last-minute consolation through Galatasaray midfielder Selcuk Inan's penalty.
USA face Germany, Portugal and Ghana in World Cup Group G.
They will meet Nigeria on 7 June in their final warm-up game ahead of their seventh consecutive appearance at a World Cup finals.
Turkey finished third at the 2002 tournament, but have not qualified since.
