Bright Osayi-Samuel put the Championship side ahead after just four minutes, scoring from the edge of the area after a swift counter attack.
Winger Pawel Wszolek then doubled the home side's lead 14 minutes later with a low strike from 20 yards.
Matt Smith's second-half header sealed victory, before Ed Upson's shot went in off Osman Kakay for Rovers.
It gave manager Steve McClaren a second successive win, after a difficult start to his reign at Loftus Road.
Saturday's win over Wigan was their first win in the Championship following four straight losses, which included a 7-1 defeat at West Brom.
