In front of a crowd of 38,707 at the Estadi Olimpic, Aitana Bonmati put the hosts ahead in the 17th minute with a low strike from 18 yards.
Barca hit the woodwork three times, but Caroline Graham Hansen made it 2-0 and Mariona Caldentey curled in a third goal before half-time.
Goals late on from Claudia Pina and Vicky Lopez sealed the emphatic win.
Barca, whose side included England internationals Lucy Bronze and Keira Walsh, have now won all nine of their league matches and are six points clear of second-placed Real.
European champions Barcelona are in action again on Wednesday when they play in Germany against Eintracht Frankfurt in the Women's Champions League.
Real, who drew 2-2 at home to Chelsea in their first Group D match last Wednesday, face Swedish side Hacken on Thursday.
