Scotland raced into a 12-0 lead but two Spanish tries levelled the game before the superior fitness and technique of Philip Doyle's side shone through.
The Scots kick off their Six Nations campaign away to Ireland on 2 February, looking to improve on their five defeats in the tournament last year.
