Ospreys let slip a first away win of the season as Benetton capped their comeback with a late try.
A stunning solo score by Max Nagy and Luke Davies' clever finish saw a much-changed Ospreys lead 13-7 in Treviso.
But Nagy suffered a serious ankle injury and two missed conversions left the door open for the hosts who remain unbeaten at Stadio Monigo since March.
Jacob Umaga levelled with two penalties before Giacomo Da Re darted over four minutes from time.
Ospreys collected a losing bonus point but remain 11th while Benetton, beaten just once in seven games this season, climbed to sixth.
Ospreys showed nine changes from the victory over Scarlets, including a first start for try-scorer Dan Edwards that last just 20 minutes.
The Wales Under-20s fly-half kicked the opening points but was forced off following a brutal introduction to URC rugby with a thunderous tackle by former All Black Malakai Fekitoa.
It meant an earlier than expected return for Wales fly-half Owen Williams following as month off with a hamstring injury.
Williams kicked well out of hand, defended well but will be disappointed with the two missed conversions.
Lock James Fender and flanker Harri Deaves carried hard, captain Morgan Morris led by example and wing Luke Morgan was lively.
In-form Benetton were knocked off their stride by the Ospreys' defence and were limited to a 7-3 lead at half-time, through hooker Bautista Bernasconi.
The Italians edged the scrums but twice the Ospreys forwards, spearheaded by debut hooker Marnus van der Merwe on loan from Cheetahs, stopped Benetton's driving lineout short of the line.
Nagy also denied opposite number Rhyno Smith with a last-ditch tackle.
Ospreys had chances of their own as Nagy's kick almost set up Iestyn Hopkins while a break by Deaves and Morris should have produced more but scrum-half Luke Davies misplaced his pass.
Nagy ecstasy and agony
Having frustrated their hosts in the first-half, Ospreys stunned Stadio Monigo into silence with a try out of nothing just 72 seconds after the break.
Nagy bumped off Umaga and then angled his 40-metre run to the corner to put Ospreys ahead 8-7.
But just four minutes later, Nagy was carried off on a stretcher after falling awkwardly for a high ball under pressure from Smith.
It was a major blow for Nagy, perhaps the in-form full-back in Wales this season, but Ospreys rallied from the second injury loss of the night.
Williams produced a wonderful 50-22 kick only for the forwards to fluff the lineout.
But when Ospreys won a surprise scrum penalty just moments later, they braved kicking to the corner again and this time Deaves pulled out a smart pass to the narrow side for scrum-half Davies to squeeze in at the corner.
However, Williams again missed the conversion, allowing Umaga to level with two penalties before Benetton patience and perseverance paid off as replacement Da Re darted into the corner to snatch victory.
Ospreys head coach Toby Booth said: "My over-riding emotion is pride with an obvious tinge of frustration. A few key moments didn't go our way to let them back in it but a lot of players will have learned from that.
"The way they [Benetton] celebrated at the end was as if they had won the World Cup so you can see how hard we pushed a team of internationals and All Blacks at home.
"We had to continually adapt. Losing Dan [Edwards] early was frustrating and than Max Nagy was a big loss. It doesn't look good for him."
