Keenan last played in April while Ryan has been out since injuring his bicep during Ireland's Six Nations campaign.
Leinster boss Leo Cullen has, however, rested several of his frontline players, with Jamison Gibson-Park, Andrew Porter, Tadhg Furlong, Josh van der Flier and Caelan Doris among those not travelling to Belfast.
With Garry Ringrose still not fit, the versatile Jimmy O'Brien starts at outside centre while flanker Will Connors starts having been a late withdrawal from the side before the big win over Ospreys last week.
Leinster are second in the URC table, a point behind leaders Glasgow, ahead of the penultimate regular-season game at Kingspan Stadium.
Ulster, meanwhile, are sixth as they bid to seal their play-off spot with a top-eight finish and, should results elsewhere go their way, a place in next season's Champions Cup.
This is Richie Murphy's first game since being appointed Ulster head coach on a two-year deal having led the team on an interim basis since March.
Ulster boosted their play-off hopes with a 31-20 win over Scarlets in Llanelli last week, while they edged Leinster 22-21 in Dublin when the sides last met in Dublin on New Year's Day.
