The M1 is likely to be shut in both directions all day between Barnsley and Hoyland following an overnight crash.
Emergency services were called to the scene between junctions 37 and 36 just after midnight.
National Highways said a car had left the carriageway in a "serious collision" and added that the closure is "expected to remain in place throughout the day".
A spokesperson added: "Due to the severity of the incident, collision and forensic investigation needs to be carried out with daylight.
"This is currently ongoing at the scene by the South Yorkshire Police collision investigations team."
Drivers have been advised to avoid the area, with police contacted for further details.
