About 80,000 people are expected to attend the festival across the weekend of 14 and 15 June.
Armand Van Helden, Rudimental, Pawsa, and Bicep presents Chroma have also been added to the line-up.
Others newly-added performers include Joy Orbison, Skream & Benga and Chris Stussy.
50 Cent joins already confirmed artists including Confidence Man, DJ Heartstring, Salute presents True Vision, and Interplanetary Criminal.
More than 100 performers will appear at this year's Parklife, which has run for 15 years despite a two-year hiatus caused by the Covid-19 pandemic.
