The 38-year-old's return, after seven years away from the WWE, was announced during the Raw programme on Monday, sparking wild reactions from fans.
Johnson, a former WWE champion and arguably one of its biggest ever stars, has appeared in a string of Hollywood films including The Scorpion King, The Mummy Returns, and The Other Guys.
The show takes place on 3 April 2011.
The Rock returned to the ring during Monday's Raw event, delivering a dramatic comeback speech which had fans chanting all his trademark slogans and catchphrases.
"Thank you, I love you and it is because of you that I am back in this ring," he announced.
'Most electrifying man'
"I give you my word, I am never, ever going away. Simply put, ladies and gentlemen, The Rock is back.
"I am back in the middle of this ring because of you," he told a frenzied audience.
He later recorded a special message for fans from his dressing room.
"Finally The Rock has come back to Wrestlemania," he told them.
"The most electrifying man in entertainment hosting the most electrifying event in entertainment," he added.
His comments have led to speculation that he may fight again, as well as raising questions about whether he is back with the sports entertainment series for good.
Wrestlemania XXVII takes place in Atlanta, Georgia - the annual show is the biggest event of the year in the WWE calendar.
