Hampshire's David Balcombe completed a career-best eight-wicket haul but Gloucestershire edged day two.
Resuming on 195-4, Gloucestershire could only add 119 runs as they were bowled out for 314 with Balcombe (8-71) taking all six remaining wickets.
Simon Katich led Hampshire's response reaching 74, before being caught by David Payne off Ian Saxelby (3-37).
But the hosts were restricted to 182-7, 132 runs behind, as Will Gidman (3-43) also excelled with the ball.
