Kent fast bowler Charlie Shreck claimed a five-wicket haul to leave Division Two leaders Derbyshire in trouble.
After Kent moved on from their overnight 238-8 to be bowled out for 265, Shreck took 5-41 as Derbyshire were skittled for just 132.
At 51-6, the home side were even in danger of following on until tail ender David Wainwright's unbeaten 29.
Kent then moved on to 93-3 second time around, Rob Key finishing unbeaten on 65 to earn a healthy 226-run lead.
