Surrey moved above Glamorgan in the South Group with victory in Cardiff.
Surrey posted 165-5 from their 20 overs after being put in to bat, Hashim Amla (55) and Kevin O'Brien (40 not out) making valuable contributions.
Kevin Pietersen, in his second T20 innings of the summer, was out caught behind off Dean Cosker for just one.
Glamorgan captain Jim Allenby (52) set up a tense finish but teenager Tom Curran (3-23) claimed career-best T20 figures to seal a 17-run win.
With six overs remaining, the Welsh side required 61 runs with six wickets in hand, but Curran removed Chris Cooke and Ben Wright in a pivotal 15th over and the hosts failed to recover, eventually finishing 148-8 from their 20 overs.
Before the match, Surrey confirmed the signing of Sri Lanka batsman Tillakaratne Dilshan to replace South Africa's newly-appointed Test captain Amla on a deal until the middle of August.
The 37-year-old is expected to make his debut against Hampshire on 27 June.
