Three wickets each for Mark Footitt and Tony Palladino ensured Derbyshire had the edge on day one against Glamorgan.
The hosts slipped to 158-6 before Chris Cooke (74) and former Derbyshire man Graham Wagg (62) put on 91 for the seventh wicket.
Footitt (3-61) and Palladino (3-34) were the pick of the visitors' bowlers as Glamorgan were dismissed for 282.
Derbyshire faced three overs before the close and reached 8-0 at stumps, trailing by 274 runs.
