Angus Robson and Ned Eckersley shared an unbroken stand of 169 as Leicestershire responded positively to Glamorgan's massive 513-9 declared.
Robson (82 not out) and Eckersley (81 not out) saw their side to 202-1.
Earlier, Will Bragg (120) only added seven to his overnight score, but Chris Cooke (48) and David Lloyd (59 not out) boosted Glamorgan's total.
Ollie Freckingham (3-111) and Atif Sheikh (3-108) were the pick of the bowlers for Leicestershire.
Centurion Bragg fell early in the day, but the runs continued to flow for Glamorgan with Lloyd collecting nine fours in his career-best score.
However, Robson and Eckersley underlined the placid nature of the pitch, collecting 20 boundaries between them, plus two sixes for Robson, after Dan Redfern had fallen to Graeme Wagg for 24 and the visitors will be hopeful of a big total of their own on day three.
Glamorgan's David Lloyd:
"A maiden first class 50 means a lot. It's been a bit up and down so far for me in the first team, but that kind of lifts something off my shoulders and hopefully I can get some more runs in the future.
"There's a bit in the wicket if you bowl it in the right place, but we didn't really seem to find the right channels.
"Hopefully if we come back and hit good areas then there'll be a bit more, and maybe a bit of cloud cover, so fingers crossed.
"There were a couple off the seamers that stayed low, and if we get a few of them, then hopefully they'll be lbw or bowled."
Leicestershire batsman Ned Eckersley:
"It's a good wicket, we saw that yesterday from their guys, and obviously it was our turn today to try and see off the new ball and then try and capitalise towards the end of the day, which we did.
"We're in a good position now, and we can start again tomorrow and try and build towards their score, but we have to be ruthless and get into the hundreds.
"Angus and I have a good chance, and we aim to do that.
"It's been a talking point in the dressing room that there have been a few hundreds in recent seasons, but no real game changers, and that's what we need to do in this game."
