Tom Curran's 5-71 helped keep promotion-chasing Surrey on terms with Derbyshire at The Oval.
Resuming on 209-5, Derbyshire slumped to 313 all out as Curran took four wickets in the first session, including that of Chesney Hughes for 95.
Tony Palladino struck twice after lunch to reduce the hosts to 48-2, but Rory Burns made 92 to steady the innings.
Zafar Ansari (21) and Gary Wilson (36) remained unbeaten as Surrey closed on 227-5, still trailing by 86 runs.
Pace bowler Curran's efforts boosted his side's hopes of securing promotion to Division One, with Surrey needing to take 15 points from the game to earn a place in the top flight next season.
The 20-year-old now has 59 wickets in Division Two this season, second only to Derbyshire seamer Mark Footitt, with 62.
Surrey batsman Rory Burns:
"We're in quite a good spot. We could have been in a better spot but we feel we're in a very good place.
"We're obviously very disappointed that we haven't capitalised on our position, but it happens sometimes."
