Defenders Ben Davies and Tom Lockyer have withdrawn from Wales' squad for Wednesday's World Cup qualifier away to Belgium because of injury.
They will also miss Saturday's friendly with Mexico and a second World Cup qualifier at home to the Czech Republic next Tuesday.
Assistant manager Robert Page said both players suffered "significant injuries" playing for their clubs.
Wales have not called up cover for either of the players.
Juventus midfielder Aaron Ramsey had already pulled out of the squad through injury on Sunday.
Page confirmed both Davies and Ramsey - part of the Wales side that beat Belgium in the 2016 European Championships - had suffered soft tissue injuries, while Lockyer had an ankle problem.
Tottenham left-back Davies, 27, is a significant blow as he is an important first-team player with 58 caps to his name.
Luton centre-back Lockyer, 26, has also featured regularly in recent seasons, earning 13 caps.
"It's not great news for us," said Page, who added Davies and Lockyer both reported for duty with the problems.
"It's not ideal preparation with only two days before the game, but it is what it is and we've got to react to that now and pick a team who we think can win us the game."
Page had initially been hopeful of Ramsey's fitness, but said a scan at his club showed the issue to be more significant than originally thought.
He added: "Of course, you want your best players and (Ramsey) certainly comes into that category; he's helped win us games in the past, important games, but it gives an opportunity to other players to shine."
Also absent is Bournemouth's David Brooks who was left out of the original selection having already been sidelined.
