Substitute Tyrese Shade's winner gave Swindon their first victory over Colchester in seven attempts.
Swindon had the ball in the Colchester net after eight minutes when Frazer Blake-Tracy powered home a header from a corner, only for it to be ruled out for a foul.
Blake-Tracy played in Luke Jephcott whose his shot was blocked, and following up, Jonny Williams then played Jephcott in behind, but Sam Hornby came out to and smother.
After 64 minutes, Swindon found the breakthrough when Williams' low ball across goal bounced off Tyrese Shade at the back post and trickled over the line.
Colchester had a chance to level as Al-Amin Kazeem found space down the left and pulled a dangerous cross across goal which Ricky Aguiar put behind.
