Aaron Hickey will miss Scotland's Euro 2024 campaign after failing to recover from a hamstring injury.
The Brentford full-back, 21, has not played for club or country since October.
Fellow right-back Nathan Patterson, of Everton, will also not make Steve Clarke's Scotland squad.
"Aaron Hickey is out, Nathan Patterson's out and we know [Lewis Ferguson] is out as well," Scotland assistant coach John Carver told BBC Radio Scotland.
"Three young guys who obviously are going to be disappointed. They have got Euros and World Cups ahead of them so hopefully we can continue to qualify and they will get their opportunity down the line."
Celtic's Anthony Ralston has also featured at right-back for Clarke, who will name a 28-man squad on Wednesday, before finalising his 26-man squad for the finals on 7 June.
Scotland will play Gibraltar (3 June) and Finland (7 June) before opening Euro 2024 against hosts Germany on 14 June.
