Newcastle recovered from a 14-point deficit to defeat Ospreys and book a place in the European Challenge Cup last eight.
Ospreys burst into the lead with tries from Keiran Williams and Dan Evans.
Yellow cards for Justin Tipuric and Adam Beard allowed Newcastle to level the half-time scores through a penalty try and score from prop Trevor Davison.
Second-half tries from Sean Robinson and Christoffel van der Walt sealed the visitors' win.
After halting a four-match losing streak, Newcastle will travel to face Leicester in the quarter-finals next weekend.
Ospreys suffered from indiscipline and missed the influence and experience of injured half-backs Rhys Webb and Stephen Myler and followed Cardiff Blues out of the European tournament.
The hosts also lost despite welcoming back six Wales internationals, with lock Alun Wyn Jones and Tipuric involved in the opening score as a flowing move was finished off by centre Williams.
Luke Price, starting in his 50th Ospreys match with Myler injured, converted.
Keelan Giles and Owen Watkin set up full-back Evans for the second score before the hosts imploded.
After failing to convert attacking opportunities because of a struggling lineout, Ospreys were reduced to 13 men with Tipuric and Beard yellow-carded.
Tipuric was penalised for a no-arms tackle by Ludovic Cayre following advice from his TMO Philippe Bonhoure and Newcastle took immediate advantage through a try from Davison.
Newcastle's penalty try came after Beard was adjudged to have collapsed the maul, an offence which saw with the Wales second-row sin-binned.
Ospreys started the second half with 13 men and Newcastle punished their hosts with a flowing move started by Callum Chick and George Wacokecoke before Robinson dived over.
Brett Connon converted to ensure Newcastle scored 21 points while Tipuric was off the field. The Ospreys skipper almost made an immediate impact on his return to the action by crossing after a searing break from Morgan Morris, but the score was ruled out by a forward pass from Price.
After another incisive attack from centre Williams, Price reduced the deficit with a penalty before another Morris surge was scuppered by a vital defensive intervention by Chick.
Newcastle hooker George McGuigan was denied a try by inches before replacement Van der Walt dived over.
Ospreys replied with a try from prop Tom Botha with Newcastle captain and England flanker Mark Wilson yellow-carded.
The hosts could not make their numerical advantage tell in the final eight minutes, though, as Newcastle celebrated victory.
Ospreys head coach Toby Booth: "We need to dust ourselves down from this. This was quite emotional for us and the changing room is absolutely devastated because this was an important competition.
"We started the game really well but we relinquished control and pressure pretty convincingly. Discipline was the basis of that and we allowed Newcastle back into the game and Premiership sides will not go away.
"We are not the team we want to be and that has just proved it. We have a lot of work to do."
Newcastle director of rugby Dean Richards: "We gave the Ospreys a bit too much respect in that opening 20 minutes and Justin Tipuric had an absolute field day in that time. They had six Welsh internationals coming back and we are not blessed with that sort of experience.
"Either side of half-time we were totally dominant and scored three tries and they had two yellow cards. I was pleased we understood how to play when they were down to 13 men and exploited it.
"We want to go as far as we can and we always value this competition."
