Flanker Jac Morgan will not play on Ospreys' two-match United Rugby Championship (URC) trip to South Africa, while hooker Dewi Lake will miss the opening game against Stormers.
Wales' 2023 World Cup co-captains have travelled with the squad but both will miss the opening game against Stormers on Saturday in Cape Town.
Lake, 24, has an outside chance of featuring against Bulls on 27 April.
Both players missed the Six Nations through injury.
Morgan, 24, has been absent since undergoing knee surgery last December, while Lake suffered a hamstring problem the following month.
On-loan Cheetahs trio Evardi Boshoff, Jeandre Rudolph and Victor Sekekete have linked back up with Ospreys this week after being unavailable for European competition.
After losing to Gloucester in the European Challenge Cup quarter-final, Ospreys are bidding for a URC play-off place by finishing in the top eight.
Toby Booth's side lie in seventh place with five regular games remaining.
