West Midlands Ambulance Service was called to the northbound carriageway near to junction 7, the Coleshill interchange, at 13:53 BST.
The driver of one of the cars, a woman, suffered serious injuries, a spokesperson said, and received specialist trauma care at the scene before being taken to Queen Elizabeth Hospital, Birmingham.
The second car had overturned and four patients - a man, woman and two children - were treated for injuries not believed to be serious before all being taken to University Hospital Coventry and Warwickshire.
