Highways England closed the stretch between junction 6 for Watlington and junction 8A for Oxford overnight while repairs took place.
The road was closed after a car towing a caravan driving the wrong way on the M40 was involved in a crash with another vehicle on Monday.
But Highways England said a vehicle had crashed between junction 5 and 6 this morning, which is continuing journey delays.

Mother Imogen gave birth to the female foal in the early hours of Friday.
Keepers say both mother and the yet-to-be-named foal are doing very well.
The latest arrival takes the total number of Grevy’s zebra at the zoo to eight and is the first foal to be sired by resident stallion Fonzy.
In the late 1970s there were 15,000 Grevy’s zebra in the wild. Today there are estimated to be around 2,800 remaining.

A pedestrian has been seriously injured in a hit and run crash in Andover.
The 48-year-old man suffered life-threatening injuries when he was hit by a motorbike in New Street shortly after 23:00 on Sunday.
The motorcyclist left the scene before police arrived.

A father who was found guilty of murdering his baby son has been jailed for life at Reading Crown Court.
Daniel McLaren will serve a minimum of 22 years in prison until he is considered for parole after he was found guilty of murdering his four-month-old baby Jack on 2 June 2017.

A former hotel on the Isle of Wight could be transformed in a multi-million-pound makeover with tree houses, woodland retreats, a restaurant, farm shop and spa.
A planning application is due to be submitted to the Isle of Wight Council to redevelop the Priory Bay Hotel
The plans include refurbishing the outbuildings to provide 14 hotel suites, a restaurant, bar and spa as well as 56 holiday lodges, 10 tree houses, 12 woodland retreats, a gym, village barn and farm shop installed.
Aria Resorts, which also owns Rookley Country Park, St Helens Holiday Park and holiday cottages in Colwell, bought the hotel, located near Bembridge, in December.
The company has come under fire after caravan owners at Rookley and St Helens were given notice to quit the sites to make way for their regeneration.
