Council leader Daren Hale said people using all the city's businesses had to behave "appropriately and responsibly".
It comes as the Euro 2020 tournament kick-off is expected to bring more fans to venues in Hull.
The easing of restrictions on 17 May allowed venues to reopen indoors as well as outside.
Rachel Stephenson, head of public protection at Hull City Council, said: "One of the biggest challenges businesses face is managing customers who fail to respect guidelines and the systems that have been put in place."
Mr Hale warned customers: "We all need to play our part" in order to keep stricter restrictions at bay.
The final stage of lifting lockdown in England is due to take place no earlier than 21 June, external. A decision is expected on Monday.
There have been 80 new coronavirus cases in Hull in the seven days up to 6 June. This is a rate of 30.8 per 100,000 of the population - an increase of 43% on the previous week.
Hull City Council said it was working with the Health and Safety Executive to visit and support local businesses.
Anyone who believes that a venue is failing to implement appropriate measures or act responsibly can contact the council's Public Protection Team, it said.
The dozen are accused of being involved in a fight at a Sheffield pub after a football derby match.

The crash, involving the biker and a Ford Fiesta, happened at about 19:55 yesterday on the A607 between Honington and Barkston, Lincolnshire Police say.
The bike rider, a man in his 50s died at the scene.
Anyone who saw what happened, or who saw the vehicles involved before the collision, is being asked to get in touch with the Lincolnshire force.
Officers say they'd also like to talk to any motorists with dash cam footage of the crash, or from around the time it happened.
A "fearsome" array of swords, machetes and knives has been seized in a raid on a group linked to about 70 knife attacks in West Yorkshire, police say.
Drugs with an estimated street value of £30,000 and £15,000 in cash were also found during the operation in the Burmantofts area of Leeds, according to West Yorkshire Police.
A total of 22 people, mostly teenage males, were arrested in the raid, the force says.
The group which was targeted has been linked to around 70 knife attacks, violent assaults, street robberies and a recent shooting in the last 12 months, officers add.
Ch Supt Damien Miller said: "The group's been behind the vast majority of incidents we've seen in the city over the last year, many of which have involved machetes being used to inflict very serious injuries."
He added: "Those involved in these incidents, both as victims and perpetrators, have often been very young teenagers."
All those arrested have been released on bail, police say.
A woman has been attacked and her car has been stolen after she used a cash machine in a South Yorkshire village in the early hours of this morning, police say.
The victim was attacked by a man as she returned to her car after using the machine in Knollbeck Lane, Brampton Brierlow, at about 04:20, according to South Yorkshire Police.
The woman was pushed to the ground before the man grabbed her keys and drove away in her blue Peugeot 206, police said.
The man is described as white and about 6ft tall. He was wearing a black hoodie and dark jeans and spoke with a Yorkshire accent, police said.
Anyone who was in the area at the time and saw or heard anything suspicious is being asked to contact police.

A planned £6.5m regeneration scheme for Scarborough’s West Pier has been launched which the borough council hopes will breathe new life into the heart of the town’s South Bay.
The aim of the scheme is to secure a "prosperous working harbour, champion fishing heritage and create a must visit destination for hospitality, leisure and events," the council claims.
Councillor Janet Jefferson said: "The harbour and pier area play a critical role in Scarborough’s local economy, with strong links to the fishing and hospitality industries.
"We want to develop a scheme which supports and helps these industries to grow, while at the same time creating a destination that becomes the focal point for leisure activity and events within South Bay."
Plans include new pier entrance kiosks, refurbishment of the listed building on the pier to include improved artists’ studios, refurbishment of the harbour offices and fish market buildings and an improved and larger cafe or restaurant space.
The cost of the Scarborough Harbour West Pier regeneration scheme is being met by £5m from the government’s Towns Fund while the council is contributing £1.5m.
It's hoped work on the scheme will start in autumn next year and be completed in the summer of 2024.
