Derbyshire's Tony Palladino took a career-best 7-53 to leave the Division Two leaders in a winning position going into the final day against Kent.
The visitors crumbled from an opening position of 93-3 to 161 all out, with skipper Rob Key ninth out for 81 having added only 18 to his overnight score.
Matt Lineker (45) and opening partner Wayne Madsen (30) began the reply well.
Usman Khawaja (43no) and Wes Durston (37no) then further reduced the target to 127 as the hosts closed on 168-2.
