Accurate bowling by Glamorgan proved too much for Derbyshire as the Welsh county gave themselves a good chance of a first Championship win since June.
All-rounders Jim Allenby (3-42) and Graham Wagg (3-56) led the way as the visitors were all out for 203.
Teenage spinner Kieran Bull took two wickets in successive balls - although his two overs cost him 35 runs.
Wayne White's 38 was the top score and he then took 4-27 as Glamorgan closed on 145-5, a lead of 224.

Derbyshire gave an undistinguished batting performance, India Test player Cheteshwar Pujara managing only seven on his debut before falling to Allenby, one of six lbw victims.
None of their top five reached 20 and the last-wicket partnership of 50 between White and Mark Footitt (26) was the highest of the innings.
Bull was hit for three sixes and three fours in his brief spell, but the 19-year-old also had David Wainwright caught at cover and Tony Palladino taken at backward square leg, only for Footitt to hit the hat-trick ball to the boundary.
Jacques Rudolph (29) and Will Bragg (22) put on 61 for the first wicket when Glamorgan batted again, but White's determined bowling effort put the match back in the balance, his victims including Allenby, who was caught behind for 43.
