Interviews with shortlisted candidates will start on Monday.
Former Barnsley, Cardiff and Southend centre-back Barker has played in both Aldershot's matches since he took charge - a 1-0 defeat at Altrincham and a 2-1 win at home to Welling.
The Hampshire side are currently 18th in the Conference table, five points above the relegation zone.
The club statement thanked supporters for their patience since Scott was dismissed, and backed Barker to continue in his role.
"Aldershot's board of directors would like to again express their confidence in, support of and gratitude to Chris in tackling the challenging double role of caretaker manager and player," it said.
