Cambridge United took over at the top of League Two with Declan Drysdale the match-winner.
Centre-back Drysdale's header 11 minutes from the end - his first goal since arriving on loan from Coventry in January - proved the difference at Rodney Parade.
Cambridge are now two points ahead of Cheltenham with four games left.
Newport remain in the play-off zone, but dropped a place to seventh, three points shy of Forest Green.
Newport manager Michael Flynn said:
"You can see why Cambridge are at the top of the table. They're a tough team to crack, they're full of leaders and have got some very good players.
"We gave them the goal - it's a bad error, but we win together and lose together. I'm not going to be singling anybody out.
"There were a lot of decisions that went against us as well today."
