Championship leaders Dundee United suffered a shock Scottish Cup exit at Queen of the South as the third-tier strugglers won on penalties after a 2-2 draw at Palmerston.
Kai Fotheringham's early opener was quickly cancelled out by Gavin Reilly before Lee Connelly put Queens ahead.
Fotheringham notched a second to send the tie into extra time but Marvin Bartley's side won the shootout 4-3.
Fellow Championship side Arbroath also suffered a third-round upset.
They were dumped out 2-1 by fourth-tier Spartans, a defeat that resulted in the departure of manager Dick Campbell.
James Craigen put the League 2 side ahead before the visitors levelled through David Gold, but Arbroath regained the lead just a minute later through Jamie Dishington with a goal that turned out to be the winner.
Aidan Fitzpatrick hit a double after Blair Alston's opener as Partick Thistle beat Queen's Park 3-0 in a Glasgow derby at Firhill.
There will be three non-league teams in Sunday night's fourth-round draw, which takes place live on Sportscene on the BBC Scotland channel (19:15 GMT).
Broomhill won 3-0 at Cumnock and Brora Rangers beat Pollok 1-0, but Buckie Thistle produced arguably the performance of the round when they defeated Broxburn Athletic on penalties despite having two men sent off in the first half of an extraordinary tie.
Broxburn took until the 81st minute to open the scoring, but Darryl McHardy forced extra time by levelling in the 98th minute.
Lewis Turner's free-kick then put Broxburn ahead in the final minute of extra time, but Buckie levelled in the second minute of time added on before triumphing 5-4 on penalties.
Dumbarton came from behind in extra time to edge a nine-goal thriller in another dramatic tie at Annan Athletic.
The Sons were 2-0 up at half-time but needed Finlay Gray's deflected 88th-minute strike to force extra time. Smith's second goal of the game put Annan ahead but Michael Ruth levelled before James Hilton's strike sealed a 5-4 victory.
Ayr left it late at Peterhead - Sean McGinty levelled in the sixth minute of added time before Fraser Bryden made it 2-1 in the 116th minute - while Inverness were also forced to wait against Cowdenbeath before David Wotherspoon and Billy Mckay netted in the final 10 minutes.
Calum Gallagher's goal was enough to earn Airdrie victory at Stranraer while Morton saw off Bo'ness 4-0 at Cappielow.
The Championship trio will be joined in the draw with fellow second-tier club Raith, who beat Dunfermline on Friday.
