Although fellow League of Ireland side Dundalk beat the Magpies of Gibraltar 3-1 on aggregate at the same stage of this competition last year, City manager Ruaidhri Higgins is taking nothing for granted against a side that finished third in their domestic league last season.
"They have had a big turnover in their squad and the conditions and heat is not what we're used to," said the Derry City boss.
"We want to try and stamp our authority on the game and come back to our home place in a strong position, that is the ultimate goal."
Magpies' goalkeeper Dayle Coleing is a former Glentoran player.
The second leg of this tie will be at Brandywell on 18 July and the prize for the winner is huge - a glamour clash in the next round against Danish Giants FC Copenhagen.

While it's all eyes on the Uefa Conference League this evening, the Champions League is already underway with Larne falling 3-0 to RFK in Latvia last night.
RFS had a goal disallowed on ten minutes before Martins Kigurs gave the hosts the lead just before the half-hour mark.
An early second-half header from Martins Balodis and a stunning Stefan Panic free-kick condemned the Inver Reds to a heavy defeat in Latvia.
The Irish Premiership champions will now face an uphill battle in the second leg at Inver Park next Wednesday.

There's a shorter trip for Irish Premiership side Crusaders, who have hopped over the Irish Sea to meet European newcomers Caernarfon Town (18:30 BST) while Linfield head north to Iceland as they come up against Stjarnan (20:00 BST).
It's the first time the Icelanders have faced a team from the Irish League so it's unknown territory for both.
