Police believe the "substantial damage" at Ancaster Valley, near Grantham, has happened in the past two weeks.
Tyres, wood and household items have been dumped there and branches have been cut from trees.
Det Con Aaron Flint said: "These sites are extremely fragile and highly vulnerable to damage.
"This is not only a mindless and selfish act of vandalism but also a serious crime against our environment."
Rachel Shaw, from Lincolnshire Wildlife Trust, added: "This is a senseless act of vandalism to a wonderful nature reserve."
The area is protected under the Wildlife and Countryside Act, meaning an unlimited fine can be handed to anyone causing intentional damage.
Lincolnshire Police said the penalty up until 2015 was a fine of up to £20,000 but that has now changed, giving the courts the power to give any fine.
A teenager has been treated in hospital after being stabbed in a North Yorkshire village in the early hours of Sunday morning, police say.
The 18-year-old man was stabbed in Thorpe Willoughby near Selby at about 02:00, according to North Yorkshire Police.
A 21-year-old man has been arrested in connection with the incident, police said.
A police spokeswoman said: "We recognise such an incident will have been alarming to local people, however, please be reassured the wider community were not believed to have been at any risk at any time.
"Residents may continue to see an increased police presence in the area as we continue our investigation, including carrying out house-to-house and CCTV inquiries."

A man who suffered fatal stab wounds in a fight in a Sheffield street has been named by police.
Armend Xhika (pictured), 22, died at Northern General Hospital on 13 May.
His death followed reports of two fights on Earl Marshall Road in Burngreave and Kirton Road in Pitsmoor.
Another man in his early 20s was injured during the disorder and remains in hospital.
Two men in their 30s and a man in his 20s have been arrested on suspicion of murder, police said.
A second man in his 20s arrested on suspicion of causing grievous bodily harm with intent, and a female in her 20s arrested on suspicion of assisting an offender have been released from custody, officers said.
Supt Ian Scott said: "I remain keen to hear from anyone who witnessed this incident.
"Reports suggest there were a number of residents in Earl Marshal Road who witnessed the disorder. If you saw anything at all, or know who was involved, please contact police today."
