David Mundell spoke to Glenn Campbell on BBC Scotland, shortly after winning the constituency of Dumfriesshire, Clydesdale and Tweeddale with a majority of almost 10,000.
Asked whether Theresa May should stand down, he said: "I certainly don't think so. I think she is the best person to lead us into these negotiations."
He added: "What I hope is that Nicola Sturgeon will take her plan for a divisive second independence referendum off the table and the SNP come forward in a constructive way in relation to Brexit negotiations and I hope that Labour will adopt a constructive approach.
"We have all got to work together on what are unprecedented post war negotiations."

David Mundell says Theresa May is still the best person to lead the country into Brexit negotiations.
She may recover, of course, but last night she scarcely sounded up for that fight.
She entered this contest to gain a personal mandate and hoping for a landslide.
She has achieved neither of those aims.

Polling expert Professor John Curtice, of the University of Strathclyde, says the exit poll got the result "almost right".
It predicted that the Conseratives would win 319 seats UK-wide - they are set to win 318.
And it suggested the SNP would lose 22 seats.
"The exit poll is about as good as you're going to get," he said.
"I think we did tell the nation at 10 o'clock what the story was and the prime minister's decision to call the general election had backfired spectacularly."

Third recount in North East Fife looks like it is coming shortly. If it is not decisive a fresh counting team is likely.
May facing calls to quit after "gamble backfires"
Just one of Scotland's 59 seats remains. North East Fife is on its third recount.
The SNP are currently on 34 seats - a majority of the 59 Scottish constituencies but a drop of 21 from the 56 they won in 2015.
The Tories have gone from one seat to 13.
Labour has seven Scottish MPs and the Lib Dems have four (or possibly five if they manage to win the North East Fife recount)
Because the SNP had so much to lose the big news of the night is the SNP casualties:

It seems the Liberal Democrats were up on 1st NE Fife Count, SNP on second count. Could end in a coin toss.
Tossing coin can only happen if votes are equal. NE Fife vote was +3 Lib Dems then +2 SNP. 3rd result imminent.
