Health Secretary Vaughan Gething tells BBC Wales there is a mistrust in politicians and "an anti-politics feeling".
"We need to understand that, listen and act."
Lord Hain, manager of the Welsh Labour In campaign, has reacted to the Blaenau Gwent result by tweeting, external that it "demonstrates the extent of left-behind alienation in South Wales Valleys at economic and social prospects".
He later added that "economic alienation is deep right across Wales in Labour areas & similarly England too".
Blaenau Gwent AM and Welsh Government minister Alun Davies agreed, saying: "This is an anti-politics and anti-establishment vote. People feel alienated and disconnected."
Plaid Cymru AM not surprised at strength of Leave vote.
