Florida Governor Rick Scott has called for a nationwide moment of silence at 18:00 (22:00 GMT).
The FBI spokesman confirmed Mateen made a call to emergency services - 911 in the US - before the shooting but would not discuss the contents of the call.
Mateen purchased multiple guns in the past few days, the FBI spokesman said.
When asked why Mateen was able to purchase a gun given the fact he was on the FBI radar for terror links, the spokesman answer he was able to do so because the investigations were closed.
In a press conference, an FBI spokesman says the agency first interviewed Omar Mateen in 2013 after he made "inflammatory remarks" to a colleague. The investigation was closed.
He added that the FBI had interviewed Mateen again in 2014 about a potential connection with suicide bomber Moner Abu Salha.
The FBI investigation found no "substantial relationship" between Mateen and Salah.

The news agency of the so-called Islamic State, Amaq, claims IS "fighter" carried out the Orlando shooting, BBC Monitoring's Mina al-Lami reports.
In his earlier statement, President Obama said the US was still investigating any "sympathies" or "associations" the suspect may had had.
It is unclear if the killer had any direct links to the group.

Pulse nightclub is back tweeting after the attacks. It used its Facebook page during the shooting, telling people to get out of the nightclub and "keep running".

Mayor Eric Garcetti confirmed the LA Times, external report that a man with weapons and possible explosives had been arrested last night who said he was going to the city's Pride parade.
Garcetti said they believed the arrest was "completely unrelated" to the Orlando shooting.
NBC News is quoting sources as saying that the suspected attacker called 911 moments before shooting and pledged allegiance to leader of the so-called Islamic State.
Pope Francis has expressed deep "horror and condemnation" of "homicidal folly" in the Orlando shooting.

Florida Senator Bill Nelson released a statement on the shooting, adding that authorities have told him that there appeared to be a link to Islamic State.
“Our thoughts and prayers go out to the families of all the victims of this horrific attack in Orlando. I have spoken with the FBI and there appears to be a link to Islamic radicalism. I have conferred with additional sources and they think there is likely a connection with ISIS (Islamic State)," Sen Nelson said.
"So as we all mourn the senseless loss of life and investigators continue to learn more, we must remain vigilant and remember that if you see something, say something.”

Security is being beefed up for a LGBT festival in Washington DC, after the Orlando attack.
DC Mayor Muriel Bowser said in a statement that she has been briefed by police chief Cathy Lanier about increased security measures ahead of Sunday's Capital Pride Festival.
Bowser said Washingtonians "will not be deterred by hate as we gather to celebrate love".
