The West Midlands regional secretary of the Unite union is to try and overturn the election which led to Len McCluskey being reappointed as its leader.
The defeated candidate in the contest, Gerard Coyne, says his supporters suffered bullying and intimidation.
The trade union regulator will be asked to rule that Unite's recent general secretary election, which Mr McCluskey won by 5,500 votes, was invalid.
A spokeswoman for Unite said its members would be deeply disappointed by the timing of the announcement.

A London Midland engine named after the former Watford and England manager, Graham Taylor, will be unveiled today.
One of the company’s Class 350 models has been fitted with a commemorative plaque, which will remain for the rest of its working life.
A ceremony will be held at 12:00 on platform 10 of Watford Junction station, which will include speeches and music from a string quartet.
But the company has warned that members of the public may be refused entry to the platform if numbers exceed safe levels.
The train will be used on the West Coast Main Line and pass through stations serving many places where Taylor was manager including Watford, Aston Villa, Wolverhampton and Wembley.
Steve Helfet, head of West Coast services at London Midland, said: "We hope that passengers travelling on this train will take a few moments to remember Graham, a true football hero."
Scott Duxbury, chairman at Watford Football Club, said: "London Midland’s respectful suggestion about Graham was put to his family for approval and we’re delighted they accepted this offer.
"Alongside other planned tributes, this gesture is a mark of just how widely Graham’s work and warm personality were appreciated across our town."

Police say they believe four masked men - rather than the trio they suggested earlier this afternoon - robbed a bank in Birmingham.
Officers were called to reports of a robbery at Halifax on Alcester Road South, Kings Heath, just after 10:00.
Police say staff were threatened with bats and "a quantity of cash" was stolen.

The family of a teenage boy electrocuted at railway tracks has been left devastated, according to transport police who are investigating the circumstances surrounding his injuries and those of another boy, also electrocuted.
This morning, paramedics were called to a park near a stretch of railway in Wednesbury between Bescot Stadium and Tame Bridge Parkway.
There they found two seriously hurt teenagers, both believed to be 13 years old. They have been taken to Birmingham Children's Hospital where one is described as being in a critical condition. The other's condition is said to be serious.
Acting Insp Jacqueline Wilson, from British Transport Police, said an investigation was under way and did not wish to speculate on the chain of events.
She warned children to stay away from railway tracks and urged parents to remind them of the dangers.
"These are real trains, real lives and railways are not playgrounds."

The 33-year-old Scotland captain made 97 appearances for the Baggies after joining from Manchester United in 2015.

Fletcher will officially join Mark Hughes' side on 1 July when his contract at The Hawthorns expires.
A teenager who went on the run after being charged with attempted robberies has been sentenced to four years in a young offenders institution.
Morgan Helsby, 19, was with two teenagers when they began a crime spree on a Redditch-bound train in March 2015.
They surrounded and threatened a 20-year-old man, demanding he hand over his phone, which he refused to do.

After getting off the train in Redditch, they ordered two more men - one at knifepoint - to give up their watches and phones outside a supermarket. When they refused, one of the victims was punched in the face.
Helsby, from Arthur Street in Birmingham, failed to appear for trial earlier this year but reported to a police station in the city on 3 May.
He was sentenced at Worcester Crown Court on 25 May after admitting two attempted robberies, ABH and failing to surrender to custody.
In April, the other teens received a four and a three-year sentence in a young offenders institution.
