There has been an outbreak of coronavirus at HMP Stocken in Rutland, the prison has confirmed.
The category C men's prison, which has more than 950 inmates, said it had found positive cases on four of its seven wings.
"Our priority now is to contain the virus on infected wings, protect our shielders, vaccinate and to prevent an outbreak in the rest of the prison," it tweeted, external.
The prison said staff had "worked tirelessly" to keep the facility infection free for 11 months, but the new variant had "created significant challenges".
It added: "We appreciate this is a worrying time for families.
"We have a plan and will continue to keep everyone updated on our social media feed."

The A38 has reopened southbound in Derby after a crash at the Palm Court Island.
Highways England reported a lorry carrying 35 tonnes of bottled water overturned just before 11:00.
The clear-up has taken several hours with motorists being diverted around the scene.
Derbyshire Police said the driver only received minor injuries.

A fire at a Mansfield surgery is being investigated after it forced GPs to postpone hundreds of appointments.
Roundwood Surgery in Wood Street said on Facebook they believed it was caused by someone breaking in, although managers said it was still too early to be sure.
Today roughly 400 appointments have had to be postponed and staff are using rooms at other surgeries for emergency appointments.
Dr Milind Tadpatrikar said he has not been able enter the building to see the full extent of the fire but the main damage was to "patient care".
Police said no-one was injured and they have "not yet established" whether the fire was suspicious.
Anyone with information is asked to come forward.

A man was taken to hospital after he was attacked in a Leicester street with a machete and slashed in the back.
Leicestershire Police said the 22-year-old was targeted on Sunday in Belvoir Street after visiting a takeaway at about 23:50.
The force said the man had left the premises with another man, and while they were talking outside, he was attacked with a machete.
The suspect then ran away along Belvoir Street towards Welford Road.
The victim was treated at Nottingham's QMC and has since been discharged.
Police said there were a number of people inside the takeaway at the time and urged them to come forward.
There are fears heavy rain later could combine with melting snow to cause flooding.

Fly-tipped asbestos has been found in two parts of Leicestershire.
Hinckley and Bosworth Borough Council said the toxic waste was found on Veros Lane, Nailstone, on Friday and on Green Lane, Odstone, on Saturday.
A specialist crew will be needed to safely remove the waste.
The authority is asking for anyone with any information on who illegally dumped the asbestos to get in touch.

Warwickshire Police said the victim, a man in his 60s from Nottinghamshire, had responded to a text proclaiming to be from HSBC containing a link leading him to a site where he was told he needed to transfer funds to another account.
He went on to transfer £9,350 into an account set up by a scammer, police said, after being told his account was in danger.
The suspect has since been released under investigation, police said.

Hundreds of candles were lit outside Nottingham's Council House last night in memory of 500 people in the city who have died with Covid-19.
The city council said the unhappy milestone had been reached last week, at roughly the same time as the nation's death toll had passed 100,000.
The city centre tribute featured 500 candles to mark each life lost.
Council leader David Mellen said: "This is a tragic milestone for our city.
"It is hard to put into words how much of a toll this virus is taking on our lives – the impact of each and every one of these deaths is devastating."

Northern parts of Derbyshire have already seen snow overnight and a Met Office amber warning, external for further winter shows remains in place until 13:00.
But it doesn't end there - Derbyshire also has a yellow warning for rain on the way, from 18:00 to 04:00 tomorrow.
The Met Office said heavy showers combined with melting snow could lead to flooding.

The M1 northbound was closed this morning at junction 29, following a crash involving five cars.
Highways England said there were delays of 30 minutes but it was aiming to have the road fully open by 10:00.
It has said two lanes have now been reopened but congestion remains.
It's not yet know if there have been any injuries.

The authority said all primary and secondary routes had been gritted and conditions were expected to improve through the morning.
Four schools have also closed: Edale Church of England Primary School, Hope Primary School, Cutthorpe Primary School and St Giles Church of England Primary School in Killamarsh.
Looks like some areas of the East Midlands have had a fresh batch of snow overnight - our Weather Watchers have reported seeing the white stuff in Shirebrook, Worksop and Mansfield (pictured).
Further sleet and snow are expected this morning but most parts will just see rain and strong winds.
A drier afternoon is then anticipated with heavy cloud.
