The first of the day's three Group Ones and Quadrilateral can make up for disappointment in the 1,000 Guineas.
On that occasion, the Roger Charlton-trained filly went off as favourite but paid the price for running too keenly to the start and in the early stages of the race.
She finished third behind Love and Cloak of Spirits, who runs again here, but can get back to winning ways with jockey Jason Watson.
Nando Parrado (150-1) wins Coventry Stakes
Nando Parrado's jockey Adam Kirby
"It's great to win - full credit to Clive.
"He is tough and hung in there. He had to be brave and he is a gutsy horse.
"We liked him before he ran first time but he didn't handle Newmarket and obviously the track and ground here has helped him and he has toughed it out."
Nando Parrado wins Coventry Stakes at 150-1
Winning trainer Clive Cox
"Fair play to [owners] Paul and Marie McCartan. We were deflated after his run at Newmarket. We were expecting more from him on that day. But he came home and worked well after that.
"I couldn't be more pleased. It's not a shock but the price is a big shock.
"He is a lovely horse with a lovely pedigree and he will get a mile in due course."
Nando Parrado wins Coventry Stakes
This could be the biggest-priced winner EVER at Royal Ascot.

13:50 Coventry Stakes (6f)
And after the two-year-old fillies, now it's the time for the meeting swansong for the two-year-old colts.
Mark Johnston has three strong chances with Qaader, Army Of India and Thunder Of Niagra all stepping up in grade after winning on their respective racecourse debuts.
Jim Crowley can add to his superb week on Qaader who looked particularly impressive in his win at Newbury.
Aidan O'Brien's Ballydoyle yard has Admiral Nelson who will be relishing the stronger challenge of this race under Ryan Moore.
Campanelle wins Queen Mary
Winning jockey Frankie Dettori
"She didn't jump great out of the stalls but when you are on the outside you have the luxury of controlling it.
"Wesley is a big supporter of mine. I wish he was able to be here."
Queen Mary Stakes
Frankie is flying. Through the air on his trademark dismount after winning aboard Campanelle for American trainer Wesley Ward.
That's Royal Ascot win number 11 for Ward and a 71st triumph for Dettori.
Campanelle shares a name with a type of pasta. And Italian Dettori really is a pasta master at Ascot.

13:15 Queen Mary Stakes (5f)
This race is the final chance for the two-year-old fillies to shine at this year's Royal meeting and there are plenty of questions to answer.
There are two runners trained by American trainer Wesley Ward, Campanelle, ridden by Frankie Dettori and Royal Approval, the mount of Silvestre de Sousa.
Amazingly, Aidan O'Brien has never won this race but he has a very strong challenger in More Beautiful who already has her name in the history books after winning the first race when racing resumed in Ireland earlier this month.
It was her first run and although Ryan Moore takes the ride today in place of Seamie Heffernan, she could make it two out of two.
Keeping their distance - trainers William Haggas and Roger Varian share a laugh.
Chiefofchiefs lands Silver Wokingham
Winning jockey William Buick
"Charlie [Fellowes] was adamant that dropping back in trip would be a plus for this horse. He travelled beautifully and it was about not getting to the front too soon.
"The ground is drying all the time and is riding a bit tacky, as you'd expect, but it is better than it was yesterday."
