Essex seized the initiative against Glamorgan as they bowled the Welsh side out for just 161 in their first innings at Swansea.
Spinner Monty Panesar took 5-50, while seamer Graham Napier took 3-11, as Glamorgan collapsed on day two.
After losing Jacques Rudolph from the first ball of the day, the hosts never recovered and were all out in 60 overs.
Michael Hogan (4-38) ensured Essex did not get too far clear in their second innings, ending the day on 131-5.
They will begin day three 256 runs ahead, with Ryan ten Doeschate and Greg Smith unbeaten, after England international Panesar sparked Glamorgan's lower-order collapse.
